will ignoring a user in hotlist prevent up/down loads?

once you have placed that user in your ignore list yes he/she can not upload from you untill they log out and in resetting their user id.

Yes it will permenantly inore them unless they change their name. But one thing in that the list will stop working if it goes beyond 512 names. So you might want to keep removing the names periodicially so that they remain effective.
You can use Mxmoniter if you want to automaticially ignore leeches. A very useful tool

It willl ignore them, until such time as either.. they fall out due to the 512 entries limit, they change their name, or far more likely, their userid (last 5 numbers) changes owing to reconnecting or their secondary connection resetting, often not all that long, it's best to consider ignore entries as fairly transient short to medium term measures. MxM & Leechammer are favourite for queue control partly for this reason, if they reappear after a reconnect, these programs recheck & re ignore them if they still fail to meet the criteria you set.

I only wanted to stop the automatic message sending bots, getting files or sending them was not my intention to ignore, so I'll just do the automatic ignore of messages with mine eyes.

you can change your privacy settings in Settings > Incoming Msgs & Privacy to only allow users to PM you if either they are in your queue or you are in theirs
